Protein Precipitation Microplate
 Protocol  Ordering Information

Laboratory sample cleanup is a necessary part for analytical preparation analysis. The removal of Contaminants such as proteins, cell debris and other materials is an important step. Typically this has been done by using Acetonitrile and then Centrifugation to pellet the debris leaving the clean supernant. After this process supernatant can be used for further analysis by HPLC, GC, MS and other analysis tandem methods.  Protein Precipitation Plate, a collection plate (in a choice of two sizes), and a vacuum manifold.

The Protein Precipitation plate is a 2ml 96 well rigid polypropylene microplate. The use of polypropylene enhances the plate rigidity and chemical resistance. The plate contains a specially formulated dual membrane and has drip directors on the base for accurate dispensing of the supernatant into the paired collection plate. The dual membrane consists of two distinct layers. The top layer acts as a pre-filter removing coarse particulates.

The bottom layer has two functions:

  • Oleophobic - for retaining the well contents without dripping.
  • Fine filter - for removing fine particulate matter (0.7um) only when a vacuum is applied. A choice of collection plates is available, in 750µl or 2000µl capacities.
  • Both have wells with round bottoms which allow you excellent sample recovery.

  •   The Vacuum Manifold is a universal filter/collection manifold that can be used with plates from 350µl to 10,000µl. It comes complete with a vacuum gauge for accurate
monitoring and reproducibility, a vacuum regulator and a two way control valve. Also available is the Vacuum Assist Frame which ensures an even vacuum across all of the 96 wells.

    Typical protocol

    1) Add up to 200ul serum to each well
    2) Add 2 x V/V acetonitrile to each well
    3) Position the plate above the Well Plate (which you have already placed in Vacuum Manifold)
    4) Position the Vacuum Assist frame on top of the microplate
    5) Apply vacuum
    6) Clean supernatant collects in Well Plate
    7) Transfer plate to auto-sampler or manually inject.

96 samples can be processed at the same time with no further pipetting steps or centrifugation, they are then clean enough to inject into the most sensitive of analyte mass analysis detection systems.
