Best Method for Liquid Loading:
1. Don't Equilibrate your SINGLE StEP™ Columns .
2. Add compound (using glass pipette, etc.), spreading it as evenly as
possible on the top frit.
3. Screw down the cap tightly and apply air pressure from a disposable
plastic syringe (60mL) until compound is fully loaded onto the silica
beneath the frit.
4. Run 2-3 column volumes of your most non-polar solvent (1g of
silica=2mL) . This allows for polar solvents to be pushed out of the
column, and your compound to start with as low polarity as possible.
6. Replace the SINGLE StEP™ Column on the SINGLE StEP™ Pump, or Biotage®
and Isco® machines, and start running.

Best Method for Dry Loading:
1. Don't Equilibrate your SINGLE StEP™ Columns .
4. Add compound by pouring the silica or powder onto the frit.
5. Place a Head Space Frit on top of the powder and then press it down
(to make a frit "sandwich").
6. Place the column on the SINGLE StEP™ Pump, or Biotage® and Isco®
machines, and start running.

Isco® Machine Users please note:
You must place SINGLE StEP™ Columns in the "Face Forward" position in
order for them to fit on your machine.
